Viruses have a global impact on ecosystem ecology through mortality, nutrient cycling, and transferring genes from one host to another, yet their study is limited by complex methodologies with little validation. Here, we compare four combinations of methods currently used to concentrate (Tangential Flow Filtration (TFF) or FeCl3-precipitation) and purify (DNase only, DNase+CsCl, or DNase+Sucrose) aquatic viral concentrates. We evaluated these methods based on community composition and protein diversity using triplicate metagenomes generated from each method derived from the same ocean sample.
